Design Notes
This device is a transcriptional unit designed specifically for S. cerevisiae. It contains the Gal1 inducible promoter with Kozak sequence and the ADH1 terminator, both quite efficient at starting and terminating the transcription of long ORFs. Under the control of the Gal1 promoter, we can induce the synthesis of olivetolic acid at specific cell densities and growth phases, allowing for optimizations in several steps. Furthermore, part BBa_K5007000 was designed as an interchangeable part, capable of being switched between plasmids and assembled with other promoter-terminator pairs, optimized for different chassis and further elaborated.
